Can titles be region restricted when buying from amazon or nuuvem??

Amazon, yes. However, it isn't common.

Both yes.
And Nuuvem has better chance at selling region locked games.
There's not many games that are locked just for US. Brazil slowly becomes new Russia, which means more and more games will get region locked - if you see Brasil in name, it's 99% locked (like Saints Row 4).
