yeah, misread the topic as only villains first. and it's tough to come up with a really interesting "badass" villain from a game -- Seph's just the first one that seemed appropriate and reading through the other answers here, I'm wondering why there aren't more great villains in games.

Villians are always tougher to characterise than protaganists. Protaganists can vary from anti-hero to true hero and their motives are always easily explainable, but with villians, the more memorable ones are usually the ones out to destroy the world or something (how many remember Larson from the first Tomb Raider? Very few, because he was just an in-the-way type villian) and it's a lot harder to come up with a reasonable motivation or personality for that character that isn't "psychotic lunatic compensating for something."

Nah, it's not a lot harder to characterize a villain. Actually it's easier for the developers, as the character of the villain is not bound to the player. Try to characterize Mario, Zeldas Link, Sonic, Gordon Freeman, Chell, those nameless Doom/Quake/Shooter avatars, yourself in RTS like C&C, Warcraft etc - all those games let you easier characterize the villains, as your own role should be defined by yourself.
There are some very few exceptions like Adventures in general and maybe character driven games like Max Payne.
I think most villains just fall short, as they seem like boring stereotypes or hyped copies from other franchises (especially when I look at the OP video). Few gamestudios really focus on character but on coolness and actual gameplay (again the OP video comes to mind), which isn't always a bad thing.

Wait, Wesker's bullet time bullshit was in the games? I thought that was just something the film added.
As for my favourite hero, it's going to have to be Elena Cousland, my Dragon Age: Origins Warden. The crazy girl's a massive DPS/tank who tries to solo anything she can find, with 62% of the party damage belonging to her and her two swords.
