what if the giveaway creator already give the game but the winner not clicking the receive button?

Try contacting the winner and let them know the key is sent, so they should redeem the game and mark as received.
If the winner is not responding after the contact (after 7 days of giveaway ending) then you can request a new winner.

Did the winner activate the game on their account?

Based on my experience all my "winner cannot be contacted" situations are they didn't use the key. However if they did use the key and the game is not in their account, I would contact wherever I purchased the key and tell them to replace the key and revoke the old one.

The winner has a week to activate and mark the giveaway. You can contact them before that and ask them to activate and mark, though.
If, after a week, they don't -- you can request a re-roll.

first check their "last online" time. they may not of even been online since they won to know or redeem. if they haven't then it's safe to re-roll after 7 days. if they have then check their library and see if the won game is inside of the library, if it's there then fill out a support ticket to force feedback, if it's not then well frankly it's still safer to submit a support ticket to force feedback.

From FAQ:

The winner has redeemed my gift, but not marked it received. How do I proceed?

If your gift has been activated or redeemed by the winner, and they have not left feedback after seven days, please create a support ticket to request received feedback. A support member will review and ticket, and if the gift is activated on the winner's Steam account, approve the ticket and update the feedback to received.

File a ticket with support to let them change the feedback, but be aware that you might also have to provide some evidence of you sending the key beforehand.
