So, I have a bunch of keys and I've just had my first person who couldn't redeem due to a key being locked which I was unaware of.
I simply have them all in a notepad file and don't even remember where I got them so I don't know what's locked and what isn't so really I don't even know what to select when making a giveaway.

HOWEVER, I'm pretty sure most of them came from Humble Bundles so does anyone know which regions they lock? I've searched Google and looked around their site and can't seem to find any info on it.

You cannot gain any Info for a steam key without trying to redeem it.

Unfortunately, there's no way of knowing a key's region lock without trying to redeem it.
But you can check SteamDB to know if a game has regional restrictions.

Check Rachel's bundle threads to figure out if they have region lock, and then check steamdb to figure out what it's locked to, by matching to the sub for your region.

If you don't know what bundle it's from.. if steamdb doesn't list any subs with restrictions, you ought be safe, otherwise may be best to assume a locked-copy.

Such a pain in the ass. Why don't they just have a "US Only" option? I know they work in the US.

That has been suggested and users are advised to make a post to invite US users to their GA while whitelisting all posters to make GA exclusive.

Also while there is no way to know if you already had the key, there is a way to know "before" buying the key, which is: SteamLocks (does SteamDB for you).
