I would say if your more of an old school gamer go for Duke, if you appreciate graphics and so on go for Sam.

I don't really know about the story lines though.

I havent played their games that much. I find Duke Nukem to kind of vulgar and his humour to be kind of in your face so on that note I'd probably go for Serious Sam.

If you mean the characters? Serious Sam.

If you mean DNF vs. SS3BFE, that's more difficult. Serious Sam 3 has better graphics and has you fighting tons of enemies at once, and it's a lot of fun. Duke Nukem Forever has a lot of variety, though, and it's also a lot of fun. They've both got lots of weird and badass weapons. Both of them have some good spooky in-the-dark horrory segments, too, and both allow you to tear out cyclopean eyes. Basically, if you just want to shoot shittons of increasingly ridiculous enemies with lots of guns and occasionally smash/tear them apart with your sledgehammer and hands like the blood-craving badass he-man that you are, then Serious Sam's for you. If you want smaller battles laced with puzzles, vehicles, and tits, and to maybe draw a penis in some kid's copy of your book like the humorously juvenile he-man that you are, then it's Duke Nukem all the way. Choose your destiny.

Which game is better? I hate saying it so many times, but Duke Nukem 3D and Serious Sam are completely different. Duke Nukem had more open levels with secrets and finding things like switches and keycards
while Serious Sam has a million monsters fearless just running to you and killing you
